Breathing Rhythm Analysis

Breathing rhythm analysis is the quantitative study of the timing, pattern, and variability of inhalation and exhalation, providing insight into how respiration is regulated. It typically measures airflow, chest or abdominal movement, and respiratory timing, then compares cycle duration, breathing rate, and phase relationships across conditions or behavioral states. In neuroscience, these measurements help link respiratory rhythms to brain activity, autonomic regulation, arousal, and coordinated behaviors such as speech and movement. Analysis of altered breathing patterns can also support research on neurological and psychiatric disorders, stress responses, sleep, and the neural circuits that generate and modulate rhythmic behavior.

Circadian rhythms in voluntary wheel-running activity in mammals are tightly coupled to the molecular oscillations of a master clock in the brain. As such, these daily rhythms in behavior can be used to study the influence of genetic, pharmacological, and environmental factors on the functioning of this circadian clock.

We recently identified a novel Drosophila circadian output, temperature preference rhythm (TPR), in which the preferred temperature in flies rises during the day and falls during the night. TPR is regulated independently from another circadian output, locomotor activity. Here we describe the design and analysis of TPR in Drosophila.
