Cell Counting Kit 8

Cell Counting Kit 8 (CCK-8) is a colorimetric assay for estimating cell viability and proliferation by measuring metabolic activity in cultured cells. Its water-soluble tetrazolium salt, WST-8, is reduced by cellular dehydrogenases in metabolically active cells to produce an orange formazan dye, whose absorbance at 450 nm generally correlates with the number of viable cells. In neuroscience research, CCK-8 helps assess neuronal and glial cell survival, proliferation, and responses to drugs, toxins, oxidative stress, or injury-related conditions. The assay supports rapid, nonradioactive comparisons of treatment effects, although metabolic changes can influence readings independently of cell number.

This video case report demonstrates the unique management of a duodenal gastrointestinal stromal tumor with a rare, understudied, exon 8 KIT mutation. Neoadjuvant therapy resulted in a 30% reduction in tumor size, making the patient a candidate for a robotic-assisted duodenal sleeve resection instead of the more morbid pancreaticoduodenectomy.
