Confocal Ophthalmoscopy

Confocal ophthalmoscopy is an optical imaging technique that produces high-resolution, depth-resolved images of the retina and optic nerve, making it valuable for studying visual-system structure and disease. A focused light beam scans the tissue while a pinhole aperture blocks out-of-focus light, improving contrast and enabling optical sectioning of retinal layers. In neuroscience, this method supports analysis of retinal neurons, nerve fiber layers, and microvascular changes in living eyes. Repeated imaging can reveal structural changes over time, helping researchers investigate neurodegeneration, glaucoma, retinal disease, and relationships between ocular biomarkers and broader nervous-system function.

Photoacoustic ophthalmology (PAOM), an optical-absorption-based imaging modality, provides the complementary evaluation of the retina to the currently available ophthalmic imaging technologies. We report the using of PAOM integrated with spectral-domain optical coherence tomography (SD-OCT) for simultaneous multimodal retinal imaging in rats.

The complete construction of a custom, real-time confocal scanning imaging system is described. This system, which can be readily used for video-rate microscopy and microendoscopy, allows for an array of imaging geometries and applications not accessible using standard commercial confocal systems, at a fraction of the cost.
