Germinal Zone Transduction

Germinal zone transduction is the targeted delivery of genetic material into proliferative neural stem and progenitor cells within regions such as the ventricular and subventricular zones, enabling researchers to study how the nervous system develops. It typically uses viral vectors or in utero electroporation to introduce a reporter or transgene, which is then expressed by transduced cells and, in some cases, inherited by their descendants as progenitors divide and differentiate. This approach supports cell labeling, lineage tracing, and manipulation of neurogenesis, providing insight into neuronal migration, circuit formation, brain development, and disease mechanisms.

The ability of embryonic germinal cells to differentiate into primordial germinal cells during early development stages is a perfect model to address our hypothesis about cancer and infertility. This protocol shows how to isolate primordial germinal cells from developing gonads in 10.5-11.5 days post coitum mouse embryos.

Source: Francis, M. B., et al. Detecting Cortex Fragments During Bacterial Spore Germination. J. Vis. Exp. (2016).This video demonstrates a colorimetric method to detect cortex degradation during bacterial spore germination by measuring the release of reducing sugars over time. Using N-acetylglucosamine standards and time-point samples, the assay quantifies sugar concentration through a purple chromogenic reaction, indicating the extent of cortex hydrolysis.
