Stereotactic Brain Injection

Stereotactic brain injection is a precision technique that delivers a substance to a defined brain region using three-dimensional coordinates, making it valuable for studying neural circuits and brain function. A stereotactic apparatus stabilizes the animal’s head, while imaging or an anatomical atlas guides a fine needle or cannula to the target; controlled infusion then deposits drugs, tracers, viral vectors, or cells at a selected depth and location. In neuroscience, this method supports localized manipulation, labeling, and monitoring of neuronal populations. Its spatial precision helps investigators link molecular or cellular changes with behavior, disease mechanisms, and potential therapeutic strategies.

MicroRNAs have significant roles in brain structure and function. Here we describe a method to enforce hippocampal miRNA over-expression using stereotactic injection of an engineered miRNA-expressing lentivirus. This approach can serve as a relatively rapid way to assess the in vivo effects of over-expressed miRNAs in specific brain regions.
