Hypotonic Medium Incubation

Hypotonic medium incubation is a laboratory technique in which cells or isolated nuclei are temporarily exposed to a solution with lower osmolarity than their interior, promoting controlled swelling. Water enters through the plasma membrane by osmosis, increasing cell volume and loosening intracellular structures; carefully timed exposure helps limit membrane rupture and preserve material for analysis. In neuroscience, this approach can support neuronal and glial cell preparation, nuclear isolation, and cytogenetic examination by improving access to chromosomes or other internal components. Its effectiveness depends on medium composition, incubation duration, temperature, and the biological state of the sample.

By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.
