Incremental Current Injection

Incremental current injection is an electrophysiological technique used to measure how excitable a neuron is by applying progressively larger currents through a recording electrode. During whole-cell patch-clamp recording, the experimenter delivers stepwise current pulses while monitoring membrane voltage; increasing depolarization can reveal the transition from passive voltage changes to action potential generation and quantify features such as rheobase and firing frequency. In neuroscience, this method helps construct current-voltage and frequency-current relationships, compare intrinsic excitability across neurons or experimental conditions, and assess effects of synaptic modulation, pharmacological agents, injury, or disease.

A New Workflow for Sampling and Digitizing Increment Cores

We present a protocol to use 3D-printed mounts to fix increment cores in the field without the necessity of unpacking and gluing them on wooden mounts. The new GSC holder allows placing the cores in a core microtome to cut their surface and directly transfer them to digital image capturing.
