Modified Coronal Slices

Modified coronal slices are ex vivo brain-tissue preparations cut in the coronal plane and adapted from standard sectioning methods to preserve or expose neural circuits for study. During preparation, adjustments to cutting orientation, section thickness, tissue handling, or the retained anatomical region can improve access to targeted structures while maintaining local cellular organization and synaptic connections. In neuroscience, these slices support electrophysiology, microscopy, pharmacological testing, and circuit analysis under controlled laboratory conditions, linking cellular activity with anatomy. They enable reproducible investigations of defined brain regions while reducing the complexity of experiments in an intact brain.

This video demonstrates a modified roller tube method for the long-term culturing of brain slices from a rodent model. First, the brain slices are attached to coverslips. Then, flat-sided roller tubes with a hole on the flat side are taken, and the coverslips are positioned on the hole using adhesive discs.
