Mouse Glioblastoma Model

A mouse glioblastoma model is an experimental system that reproduces key features of glioblastoma, an aggressive primary brain tumor, in a living mammal for neuroscience and cancer research. Researchers typically implant mouse or human glioma cells into the brain, creating orthotopic syngeneic tumors in immune-competent mice or xenografts in immunodeficient mice; tumor growth then reflects interactions with brain tissue, blood vessels, and, when preserved, the immune microenvironment. These models support studies of tumor invasion, molecular mechanisms, treatment response, and recurrence, while enabling evaluation of surgery, radiation, immunotherapy, and targeted drugs before clinical testing.

Stem cells are promising therapeutic carriers to treat brain tumors due to their intrinsic tumor tropism. Non-invasive intranasal stem cell delivery bypasses the blood brain barrier and demonstrates strong potential for clinical translation. This article summarizes the basic principles of intranasal stem cell delivery in a mouse model of glioma.

Begin with a mouse model of glioblastoma, an invasive brain tumor. At the tumor periphery, the blood-brain barrier (BBB) remains intact, with endothelial cells forming tight junctions that regulate molecular entry. The mouse is fitted with a head-mounted optical fiber to deliver laser light.Administer a low-intensity infrared laser, which penetrates the skin and skull to reach the brain. The laser excites molecular oxygen in brain cells, generating reactive oxygen species (ROS).The resulting...
