Olig2-egfp Reporter

The Olig2-egfp reporter is a genetic tool that uses enhanced green fluorescent protein (EGFP) to visualize cells expressing the Olig2 transcription factor, an important regulator of neural development. In this system, Olig2 regulatory sequences drive EGFP production, causing Olig2-expressing neural progenitors and their descendants to emit green fluorescence that can be detected by microscopy or cell-sorting methods. In neuroscience, the reporter supports analysis of oligodendrocyte lineage development, motor neuron formation, and neural progenitor behavior. It enables researchers to track cell fate, isolate defined populations, and evaluate how genetic or environmental signals influence nervous-system development and disease-related processes.

This paper provides a step by step guide to the fluctuation analysis technique k-Space Image Correlation Spectroscopy (kICS) for measuring diffusion coefficients of fluorescently labeled plasma membrane proteins in live mammalian cells.

Development of Leishmania Species Strains with Constitutive Expression of eGFP

2023

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.

Data Reporting and Recording

2023

Reporting and recording are crucial in data documentation. The timely, thorough, and accurate documentation of facts is essential when recording patient data. Failure to record findings during an assessment or interpretation of a problem will result in loss of information and make the patient document unreliable. The reader is left with general impressions if the information is not specific. A recording is documenting data of the individual's health information in a traceable, secure, and...

A Lentiviral Transcriptional Reporter System to Generate a Glioblastoma Stem Cell Differentiation Reporter Line

2025

This video demonstrates a technique to generate a glioblastoma stem cell reporter line to identify the differentiation of glioblastoma stem cells (GSCs) into astroglia. It outlines the steps involved in seeding GSCs, transducing them with a lentivirus encoding GFP, and determining GFP reporter expression through flow cytometry.
