Oxygenated Perfusion

Oxygenated perfusion is a technique that circulates an oxygen-rich fluid through vascularized tissue to sustain cellular metabolism and function outside normal circulation. A pump delivers controlled flow and pressure while an oxygenator transfers oxygen into the perfusate, allowing it to reach tissue through blood vessels and support gas exchange, nutrient delivery, and waste removal. In neuroscience, oxygenated perfusion helps maintain isolated brains, brain slices, and other neural preparations for ex vivo research. It supports studies of neural physiology, metabolism, injury, electrophysiology, and responses to drugs, providing experimental control while preserving tissue function.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

Here we present a protocol describing oxygenated ex situ machine perfusion of donor liver grafts. This article contains a step by step protocol to procure and prepare the liver graft for machine perfusion, prepare the perfusion fluid, prime the perfusion machine and perform oxygenated normothermic machine perfusion of the liver graft.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.
