Photoactivation Window

A photoactivation window is the time interval during which light exposure can activate a photosensitive molecule, protein, or neural probe, making it important for controlling when experimental signals occur. In neuroscience, illumination within this window delivers photons that trigger a light-dependent conformational or chemical change, such as opening an ion channel or altering a fluorescent reporter, while timing and duration influence the resulting neural response. Researchers use photoactivation windows in optogenetics and related imaging methods to link precisely timed neuronal activity with behavior, circuit function, and cellular signaling. Defining this window helps optimize stimulation protocols and interpret temporal relationships in neural systems.

Bimolecular Fluorescence Complementation-Coupled Photoactivated Localization Microscopy

2025

This video describes BiFC-PALM ― a combination of photoactivated localization microscopy and bimolecular fluorescence complementation ― to assess protein-protein interactions. BiFC involves the fusion of two fluorescent protein fragments with two interacting proteins of interest. When the two proteins interact, the fragments are brought into proximity, which allows the two parts to come together and reconstitute a functional fluorescent protein. The fluorescence of a single fluorophore is...

Source: Boyer, N. P. et. al., Imaging and Analysis of Neurofilament Transport in Excised Mouse Tibial Nerve. J. Vis. Exp. (2020) This video demonstrates the fluorescence photoactivation method to analyze neurofilament transport in myelinated axons of peripheral nerves. By tracking photoactivated green fluorophore-tagged neurofilaments, the technique provides insights into axonal transport in neurodegenerative diseases.
