Striatal Brain Slices

Striatal brain slices are thin sections of brain tissue containing the striatum, a basal ganglia region that integrates signals involved in movement, learning, motivation, and reward. Researchers prepare acute or organotypic slices from brain tissue and maintain them in oxygenated artificial cerebrospinal fluid or culture conditions, preserving local neuronal circuits and synaptic connections for experimental study. These preparations support electrophysiology, calcium imaging, pharmacology, and circuit analysis by allowing direct manipulation and measurement of striatal neurons, including medium spiny neurons. Striatal slices therefore provide a controlled model for investigating synaptic transmission, plasticity, neural circuitry, and mechanisms relevant to neurological and psychiatric disorders.

2025

This video demonstrates a protocol to evaluate the maturation and reprogramming of glial cells into neurons in a transfected mouse striatum slice. The experiment measures resting membrane potential and response to current injections using neuron-specific fluorescent markers and electrophysiological techniques. Voltage spikes during these measurements indicate successful neuronal maturation and reprogramming.

Optogenetic approaches are widely used to manipulate neural activity and assess the consequences for brain function. Here, a technique is outlined that upon in vivo expression of the optical activator Channelrhodopsin, allows for ex vivo analysis of synaptic properties of specific long range and local neural connections in fear-related circuits.

Generation of Tilted Amygdala Slices from a Mouse Brain

2025

The video demonstrates the generation of tilted amygdala slices from a mouse brain. The brain is positioned at an angle on the vibratome specimen stage to obtain slices containing the amygdala and axons from the medial prefrontal cortex that form synaptic connections with the amygdala. The brain slices are maintained at the physiological temperature to preserve the tissue for further experiments.
