Calcium Release Inhibition

Calcium release inhibition is the reduction of calcium movement from intracellular storage compartments into the cytosol, a pharmacological strategy that regulates calcium-dependent cellular activity. It works by limiting the opening or function of channels such as ryanodine receptors and inositol trisphosphate receptors on the endoplasmic or sarcoplasmic reticulum, thereby lowering cytosolic calcium signals. Because calcium controls muscle contraction, secretion, metabolism, and many signaling pathways, this mechanism can alter tissue function without necessarily blocking calcium entry across the plasma membrane. In pharmacology, calcium release inhibition supports the study and potential treatment of disorders involving excessive intracellular calcium, including abnormal contraction, secretion, or cellular injury.

We present protocols for the application of our targeted genetically-encoded calcium indicator (GECI) CatchER+ for monitoring rapid calcium transients in the endoplasmic/sarcoplasmic reticulum (ER/SR) of HEK293 and skeletal muscle C2C12 cells using real-time fluorescence microscopy. A protocol for the in situ Kd measurement and calibration is also discussed.

We developed a gene-chimeric preparation of ventral hippocampal – accumbens circuit in vitro that allows direct live imaging to analyze presynaptic mechanisms of nicotinic acetylcholine receptors (nAChRs) mediated synaptic transmission. This preparation also provides an informative approach to study the pre- and post-synaptic mechanisms of synaptic plasticity.
