Experimental Methodology

Experimental methodology is the systematic framework researchers use to test hypotheses by collecting and analyzing evidence under controlled conditions. In psychology, it typically involves manipulating an independent variable, measuring its effect on a dependent variable, and controlling potential confounding factors through procedures such as random assignment, standardized instructions, and comparison groups. Researchers use these methods to evaluate causal relationships in areas including learning, memory, perception, emotion, and social behavior. Careful experimental design, reliable measurement, ethical participant treatment, and appropriate statistical analysis strengthen the validity and reproducibility of findings, supporting theory development and evidence-based applications in clinical, educational, and organizational settings.

We describe methods to develop an experimental model of diet-induced metabolic syndrome (MetS) in rabbits using a high-fat, high-sucrose diet. Animals developed central obesity, mild hypertension, pre-diabetes, and dyslipidemia, thus reproducing the main components of human MetS. This chronic model will allow acquisition of knowledge underlying mechanisms of disease progression.

Experimental Methodology for Estimation of Local Heat Fluxes and Burning Rates in Steady Laminar Boundary Layer Diffusion Flames

We describe the use of micro-thermocouples to estimate local temperature gradients in steady laminar boundary layer diffusion flames. By extension of the Reynolds Analogy, local temperature gradients can be further used to estimate the local mass burning rates and heat fluxes in such flames with high accuracy.

The synthesis of asymmetric species of ferrocene is challenging using solution techniques. This report focuses on the methods carried out to produce a ferrocene-biotin bioconjugate using facile and clean reactions accomplished via solid-phase synthesis. Incorporation of a thiolate moiety is shown to impart the ability for immobilization on gold surfaces.

Multiplexed Fluorometric ImmunoAssay Testing Methodology and Troubleshooting

Using Luminex Corporation’s xMAP microsphere technology, we have developed the Multiplexed Fluorometric ImmunoAssay (MFIA) for serosurveillance of various laboratory animal species. The MFIA is a suspension microarray where antigen, tissue control or immunoglobulins are covalently linked to color-coded polystyrene microspheres. The MFIA testing method as well as various troubleshooting topics is addressed.
