Covalently Linked Protein Regulators

Ces modifications jouent un rôle important dans la fonction et la stabilité de ces protéines. Les molécules liées de manière covalente comprennent des groupes fonctionnels, tels que les groupes méthyle, acétyle et phosphate, ainsi que de petites protéines, telles que l'ubiquitine. Il existe environ 200 types différents de régulateurs covalents identifiés.

Ces groupes modifient des acides aminés spécifiques dans une protéine. Les groupes phosphate ne peuvent être liés de manière covalente qu'aux acides aminés sérine, thréonine et tyrosine, tandis que les groupes méthyle et acétyle ne peuvent être liés qu'à la lysine. Ces groupes sont ajoutés et supprimés d’une protéine par une enzyme ou une paire d’enzymes. Par exemple, une acétyltransférase ajoute un groupe acétyle à une protéine et une désacétylase peut le supprimer. La p53 subit diverses modifications en réponse à divers types de stress, notamment les radiations et les agents cancérigènes. Certaines modifications incluent la phosphorylation, l'acétylation et la sumoylation en réponse aux rayonnements UV et gamma. Les sites et les types de modifications peuvent varier en fonction du facteur de stress. Des études ont montré que les rayons UV et gamma peuvent entraîner la phosphorylation de la sérine 33, mais que la sérine 392 peut être phosphorylée lorsqu'elle est exposée aux rayons UV, mais pas aux rayons gamma. Proteins can be changed after they are made by adding small chemical tags or small proteins. These post-translational modifications help control protein function and stability when the cell environment changes. Common covalent regulators include methyl, acetyl, and phosphate groups, as well as small proteins such as ubiquitin.

About 200 types of covalent regulators have been identified. These tags attach to specific amino acids in a protein. Phosphate groups can attach only to serine, threonine, and tyrosine. Methyl and acetyl groups can attach only to lysine.

Enzymes add and remove these tags. For example, an acetyltransferase adds an acetyl group, and a deacetylase removes it. The effect of a tag depends on both its number and its position on the protein. A single ubiquitin on a cell surface receptor can send that protein into endocytosis. Multiple ubiquitins linked together can mark the same protein for proteolytic degradation.

A single protein can carry several modifications at the same time. One important example is p53, a tumor-suppressor protein. p53 changes in response to stress such as radiation and carcinogens, and it can receive phosphorylation, acetylation, and sumoylation. These changes can vary with the stress type, the cell type, and the organism.

UV and gamma radiation can both lead to phosphorylation of serine 33 in p53. Serine 392 can be phosphorylated after UV exposure but not after gamma radiation. Other stress conditions, including hypoxia, anti-metabolites, and actinomycin D, can cause acetylation of p53. These examples show how covalent tags help cells fine-tune protein behavior in different conditions.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
