Le matériel génétique, l’ADN, d’une cellule eucaryote existe à l’intérieur du noyau sous forme de structures linéaires appelées chromosomes.
À l’intérieur des chromosomes, l’ADN double brin s’enroule principalement autour de protéines nucléaires octamères appelées histones pour former des nucléosomes. Plusieurs protéines non-histones s’associent également à l’ADN, aidant à l’emballage de l’ADN.
Ces nucléosomes et protéines non histones s’organisent pour former des bobines qui se replient ensuite en boucles, dans un réseau, formant une chromatine emballée. L’étendue de la compaction de la chromatine permet à un ADN extrêmement long de s’insérer dans le noyau. Pendant la phase M, la chromatine s’enroule davantage pour former les chromosomes condensés.
La duplication chromosomique implique la duplication de l’ensemble de la structure de la chromatine, de sorte que l’ADN et les protéines associées à la chromatine sont tous dupliqués. La production d’histones est augmentée pour générer les protéines supplémentaires nécessaires à l’emballage de l’ADN nouvellement synthétisé en chromosomes.
La réplication de la structure chromatinienne exacte du chromosome dupliqué est essentielle à la régulation des gènes. 
Au sein des chromosomes, le niveau d’emballage de la chromatine n’est pas toujours uniforme. Les chromosomes contiennent des régions avec une chromatine étroitement tassée appelée hétérochromatine et des régions avec une chromatine faiblement tassée appelée euchromatine.
L’ADN de l’hétérochromatine est inaccessible à la machinerie de transcription et, par conséquent, les gènes de ces régions ne sont pas régulièrement transcrits. En revanche, l’ADN de l’euchromatine est plus accessible, et les gènes de ces régions peuvent donc être transcrits. 
Cette importance structurelle et fonctionnelle de la chromatine rend crucial le fait que l’ensemble de la structure de la chromatine soit reproduite avec précision lors de la duplication des chromosomes.

Duplication of Chromatin Structure

Le processus de duplication des chromosomes au cours de la division cellulaire nécessite une perturbation et un réassemblage de la chromatine à l’échelle du génome. La structure de la chromatine doit être héritée, réassemblée et maintenue avec précision dans les cellules filles pour assurer la propagation de la lignée.

L'unité de base de la chromatine est le nucléosome, constitué d'ADN enroulé autour de protéines histones octamériques et de courtes séquences d'ADN de liaison séparant les nucléosomes individuels. Les protéines histones du nucléosome ont leurs extrémités N-terminales dépassant du noyau, fournissant des sites pour diverses modifications covalentes qui régulent la structure et la fonction de la chromatine.

Au cours de la réplication, à mesure que l'ADN se déroule, les nucléosomes parentaux sont perturbés et les protéines histones sont libérées. Au fur et à mesure que la réplication progresse et que des brins filles se forment, les histones parentales et les protéines histones supplémentaires synthétisées au cours de la phase S sont assemblées, permettant la formation de nucléosomes.

Les modifications post-traductionnelles des histones et d’autres domaines épigénétiques de l’ADN sont également fidèlement reproduites dans le génome fille.

La structure de la chromatine influence l'expression des gènes 
Au sein d’une cellule, la majeure partie du génome reste inaccessible aux facteurs de transcription, car les séquences d’ADN régulatrices et codantes sont pour la plupart cachées dans les nucléosomes. Pour qu’un gène s’exprime, il est nécessaire de créer des sites accessibles pour que les facteurs de transcription se lient et également de modifier les histones pour réorganiser la structure de la chromatine et créer un environnement permissif pour la transcription.

Des complexes de facteurs régulateurs spécifiques sont impliqués dans l'ouverture de régions localisées de la chromatine par déplacement ou perturbation des nucléosomes. Les modifications post-traductionnelles de la queue de l'histone servent à maintenir un état transcriptionnel actif ou inactif. Des modifications spécifiques au niveau de la queue de l'histone peuvent atténuer le niveau de compaction de l'ADN, facilitant la déstabilisation et le déplacement des nucléosomes, donnant ainsi accès à la machinerie de transcription pour influencer l'expression des gènes.

Chromatin rebuilding during DNA replication helps cells copy DNA and pass on chromosome structure to daughter cells. During cell division, the genome must be disrupted and then reassembled across the whole chromosome. This process allows chromatin to be inherited, rebuilt, and maintained in new cells.

The basic unit of chromatin is the nucleosome. A nucleosome contains DNA wrapped around an octamer of histone proteins, with short linker DNA between neighboring nucleosomes. The histone proteins have N-terminal tails that stick out from the core. These tails can receive covalent modifications that help control chromatin structure and function.

As DNA unwinds during replication, parental nucleosomes are disrupted and histone proteins are released. When daughter DNA strands form, the old histones and the extra histone proteins made during S phase are put back together on the DNA. This assembly recreates nucleosomes on the new strands. The same histone marks and other epigenetic features in the DNA are also copied into the daughter genome.

Chromatin structure also affects gene expression. In most cells, much of the genome is not available to transcription factors because regulatory and coding DNA is hidden inside nucleosomes. To turn a gene on, the cell must open a local DNA region so transcription factors can bind. It must also modify histones to rearrange chromatin into a state that supports transcription.

Regulatory factor complexes help open these local chromatin regions by displacing or disrupting nucleosomes. Histone tail modifications can help keep chromatin in an active or inactive state. Some modifications reduce DNA compaction and make nucleosomes easier to destabilize or move. This gives the transcription machinery access to the DNA and helps control gene expression.
