Optical Redox Ratio

The optical redox ratio is a label-free fluorescence metric that estimates cellular metabolic state from the relative signals of endogenous reduced NAD(P)H and oxidized flavins such as FAD. During imaging, excitation of these metabolic cofactors produces autofluorescence, and their intensity ratio changes as electron transfer, oxidative phosphorylation, and glycolysis alter the balance between reduced and oxidized molecules. In bioengineering, optical redox ratio measurements provide noninvasive information about cell viability, differentiation, stress, and metabolic heterogeneity in engineered tissues and biomaterials. Because the method avoids exogenous labels, it can support real-time monitoring of tissue development, disease models, and responses to therapeutic or environmental conditions.
