Lipid Separation

Lipid separation is the isolation and resolution of lipids from complex biological mixtures, a key step for identifying lipid classes, measuring composition, and studying membrane and metabolic function. It works by exploiting differences in properties such as polarity, solubility, charge, and molecular size, using approaches that may combine solvent extraction with chromatographic separation, including thin-layer or liquid chromatography. In biological research, these methods help prepare samples for lipid profiling, characterize membrane components, and track changes in pathways associated with disease, nutrition, or cellular stress. Reliable separation improves analytical specificity and supports comparisons among tissues, organisms, and experimental conditions.

In reptiles, skin lipids from conspecifics are crucial for sexual signaling, with potential use in invasive species management. Here, we describe protocols for extracting skin lipids from shed skin or whole animals, determining and analyzing the total lipid mass, and separating the lipids using fractionation via column chromatography.

Dialysis: Diffusion Based Separation

2023

Dialysis is a common technique used in biochemistry for separating molecules based on diffusion. In this procedure, a semipermeable membrane allows the movement of certain molecules based on size. This method can be applied to the removal of buffer, known as desalting, or exchanging buffer molecules or ions from a protein solution. This video covers the principles of dialysis along with a general procedure. Several applications of dialysis are reviewed, including the removal of gradient...
