Protein Detergent Complexes

Protein detergent complexes are assemblies in which detergent molecules surround and solubilize hydrophobic regions of proteins, particularly membrane proteins, enabling their study in aqueous laboratory solutions. Above the detergent’s critical micelle concentration, amphipathic molecules associate with nonpolar protein surfaces to form micelle-like shells that replace the surrounding lipid bilayer while helping maintain protein solubility. In biological techniques, these complexes support membrane protein extraction, purification, electrophoresis, and structural analysis. Detergent selection and concentration can strongly influence protein stability, conformation, activity, and compatibility with downstream assays, making complex formation an important consideration in biochemical research.

Detergent Purification of Membrane Proteins

2023

Detergents are used to purify the integral proteins of the membrane. The hydrophobic portion of the detergent can replace membrane phospholipids while solubilizing the membrane proteins. When detergent monomers reach a specific concentration in a solution called critical micelle concentration (CMC), they form micelles. Above CMC, the concentration of the detergent monomers remains in equilibrium with the micelle. The number of detergent monomers present in the CMC varies for each detergent, and...

In this protocol we demonstrate the expression, solubilization, and purification of a recombinantly expressed membrane protein, MexB, as a soluble protein detergent complex. MexB is a multidrug resistance membrane transporter from the opportunistic bacterial pathogen Pseudomonas aeruginosa.

Protein Complex Assembly

2020

Proteins can form homomeric complexes with another unit of the same protein or heteromeric complexes with different types. Most protein complexes self-assemble spontaneously via ordered pathways, while some proteins need assembly factors that guide their proper assembly. Despite the crowded intracellular environment, proteins usually interact with their correct partners and form functional complexes. Many viruses self-assemble into a fully functional unit using the infected host cell to...

Protein Complexes with Interchangeable Parts

Groups of proteins may form a complex where each protein in this complex has a different role in the overall execution of the complex’s function. Often some of the proteins in the complex can be replaced by a closely related variant to give a complex that contains many of the same components yet is functionally distinct. The SCF ubiquitin ligase is a protein complex of five individual proteins. This complex attaches ubiquitin to other target proteins to mark them for degradation. In order to...
