Yfp-mcherry Reporter

The YFP-mCherry reporter is a dual-fluorescent protein construct used to visualize gene expression, protein localization, or cellular behavior in living cells. It works by producing yellow fluorescent protein (YFP) and mCherry, whose distinct excitation and emission properties allow researchers to distinguish two fluorescent signals by microscopy and compare their distribution or timing. In biological techniques, this reporter supports monitoring promoter activity, tracking tagged proteins, assessing cellular dynamics, and identifying relationships between molecular events in the same specimen. Its two-color output can improve spatial and temporal analysis while reducing the need for separate samples or labeling procedures.

Multiphoton Microscopy of Cleared Mouse Brain Expressing YFP

Multiphoton microscopy of whole mouse organs is possible by optically clearing the organ before imaging, but not all protocols preserve the fluorescent signal of fluorescent proteins. Using an optical clearing method with ethanol-based dehydration and benzyl alcohol:benzyl benzoate clearing, we show high-resolution multiphoton images of whole mouse brain expressing YFP.

The In Vitro Single Oocyte Reporter Assay: A Technique to Study Target mRNA Translation Regulation During In Vitro Oocyte Maturation

In this video, we demonstrate that in the presence of a phosphodiester inhibitor, the oocyte cell cycle is arrested at prophase-I, during which transcribed mRNAs are stored in the absence of translation. After the release of the inhibitor, oocyte maturation resumes, and the cells enter metaphase. The signal of reporter mRNA fused with the target gene in metaphase-I indicates the activation of translational machinery and protein expression.
