Bridge Slide Preparation

Bridge slide preparation is a microscopy technique that creates a shallow chamber for observing living specimens without compressing them beneath a coverslip. The method uses raised supports on the slide to hold the coverslip above the specimen, while a liquid sample occupies the space between them and maintains a moist viewing environment. This preparation is useful for examining cell shape, movement, and short-term behavior in organisms such as microorganisms or other delicate biological samples. By reducing mechanical distortion and improving specimen survival during observation, bridge slides support clearer, more representative microscopic analysis in biology.

A procedure for creating and imaging capillary bridges in slit-pore geometry is presented. The creation of capillary bridges relies on the formation of pillars to provide a directional physical and chemical heterogeneity to pin the fluid. Capillary bridges are formed and manipulated using microstages and visualized using a CCD camera.
