Cell-to-cell Contact

Cell-to-cell contact is the direct physical interaction between neighboring cells, enabling tissues to maintain structure, coordinate behavior, and exchange information. It occurs through specialized adhesion molecules and membrane junctions, including cadherin-based adherens junctions, tight junctions, gap junctions, and receptor-ligand interactions that connect cytoskeletal networks or permit the passage of ions and small molecules. These contacts regulate cell polarity, migration, proliferation, differentiation, and tissue barrier function. Studying cell-to-cell contact helps explain development, immune responses, wound repair, and disease processes such as cancer metastasis, while supporting research in tissue engineering and regenerative biology.

This protocol describes a fluorescence fluctuation spectroscopy-based approach to investigate interactions among proteins mediating cell-cell interactions, i.e. proteins localized in cell junctions, directly in living cells. We provide detailed guidelines on instrument calibration, data acquisition and analysis, including corrections to possible artefact sources.

Contact-dependent cell communication, as the name suggests, requires that communicating cells be in direct contact with each other. This is achieved either through receptor-ligand interactions or by specialized cytoplasmic channels that allow the flow of small molecules between cells.
