Commercial Kit Modification

Commercial kit modification is the adaptation of a ready-to-use biological assay or laboratory kit to meet the requirements of a particular sample, experiment, or research question. Researchers may adjust sample preparation, reagent volumes, incubation conditions, detection settings, or protocol steps while preserving the kit’s underlying biochemical principle, such as antibody binding, enzymatic reaction, nucleic acid amplification, or fluorescence detection. In biology, these modifications can improve compatibility with complex specimens, increase sensitivity, reduce reagent use, or extend a kit to new applications. Careful optimization and validation are essential because changes can affect assay performance, reproducibility, controls, and interpretation of results.

2011

A reliable and useful approach to detect histone modifications on specific plant genes is described. The approach combines chromatin immunoprecipitation (ChIP) and real-time quantitative PCR. It allows detection of histone modifications on specific genes with a role in diverse physiological processes.

Histone Modification

2020

The histone proteins have a flexible N-terminal tail extending out from the nucleosome. These histone tails are often subjected to post-translational modifications such as acetylation, methylation, phosphorylation, and ubiquitination. Particular combinations of these modifications form “histone codes” that influence the chromatin folding and tissue-specific gene expression. Acetylation The enzyme histone acetyltransferase adds acetyl group to the histones. Another enzyme, histone deacetylase,...

2009

The Protease Fluorescent Detection Kit is designed for the measurement of protease activity using fluorometry. It is also suitable for detection of trace amounts of protease contamination. The method is based on the proteolytic hydroysis of a proprietary formulation of a FITC-labeled casein substrate.

Analysis of the c-KIT Ligand Promoter Using Chromatin Immunoprecipitation

DNA-protein interactions are essential for multiple biological processes. During the evaluation of cellular functions, the analysis of DNA-protein interactions is indispensable for understanding gene regulation. Chromatin immunoprecipitation (ChIP) is a powerful tool to analyze such interactions in vivo.

The histone proteins in the nucleosomes are post-translationally modified (PTM) to increase or decrease access to DNA. The commonly observed PTMs are methylation, acetylation, phosphorylation, and ubiquitination of lysine amino acids in the histone H3 tail region. These histone modifications have specific meaning for the cell. Hence, they are called "histone code". The protein complex involved in histone modification is termed as "reader-writer" complex. Writers The writer is an enzyme that can...
