Gel Fixation

Gel fixation is a laboratory treatment that preserves molecules separated within a gel, helping maintain band position and signal during subsequent analysis. After electrophoresis, a fixative solution, often containing alcohol or acid, immobilizes or precipitates proteins and other analytes in the gel while removing interfering substances and limiting diffusion. This step supports reliable staining, imaging, and quantitative interpretation in techniques such as polyacrylamide gel electrophoresis and protein analysis. By stabilizing separated components before visualization, gel fixation improves the clarity, reproducibility, and long-term preservation of electrophoretic results in biological research.

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column.
