Neonatal Mouse Spleens

Neonatal mouse spleens are developing lymphoid organs that support blood filtration, immune-cell formation, and maturation during the early postnatal period, making them useful models in biology. After birth, the spleen undergoes rapid structural and functional changes as hematopoietic activity shifts, lymphocyte populations expand, and immune compartments become organized in response to developmental signals and environmental exposure. Researchers examine neonatal spleen tissue, cell populations, and gene expression to study immune development, hematopoiesis, infection, inflammation, and genetic disease. Findings from this model can clarify how early immune systems are established and provide a foundation for investigating developmental disorders and testing experimental interventions.

A Mouse Model of Vascularized Heterotopic Spleen Transplantation

2025

The video demonstrates a surgical method for generating a mouse model of vascularized heterotopic spleen transplantation. The abdominal cavity of an anesthetized donor mouse is surgically opened to remove the spleen. The inferior vena cava and infrarenal aorta are identified, their lumbar branches are ligated, and openings are made for the heterotopic transplantation of a spleen from a congenic donor mouse. Upon completing vascular anastomosis and restoring blood flow to the implanted spleen,...

Assessing Bacterial Burden in the Mouse Spleen for Infection Studies

Source: Madenspacher, J. H. and Fessler, M. B. A Non-invasive and Technically Non-intensive Method for Induction and Phenotyping of Experimental Bacterial Pneumonia in Mice. J. Vis. Exp. (2016)This video demonstrates the process of quantifying bacterial load in the spleen of an infected mouse to assess the severity of infection. The homogenized spleen is serially diluted and plated to determine total bacterial burden. The resulting data provide insights into host-pathogen interactions within...

We describe a protocol herein for isolating intact islets from neonatal mice. Pancreata were partially digested with collagenase, followed by washing and hand picking. 20 - 80 islet clusters can be obtained per pancreas from newly born mice, which are suitable for several islet studies.

Primary mouse cardiomyocyte cultures are one of the pivotal tools for the investigation of myofibrillar organization and function. The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The resulting cardiomyocyte cultures may be subsequently used for a variety of biomechanical, biochemical and cell-biological assays.
