Dss Dose Titration

DSS dose titration is the systematic adjustment of dextran sulfate sodium concentration to produce a controlled range of intestinal injury in experimental models, an important step for reproducible cancer research. DSS is administered at defined doses, and resulting changes in disease severity are assessed through measures such as body weight, stool characteristics, bleeding, colon length, and tissue pathology; comparing these responses establishes a dose-response relationship. This approach helps researchers select conditions that model acute or chronic colitis and investigate inflammation-driven colorectal cancer, while limiting excessive morbidity and improving consistency when evaluating disease mechanisms, prevention strategies, or therapeutics.

Experimental models of inflammatory bowel disease have allowed us to examine the complex innate and adaptive immune responses associated with pathogenesis. Using histological scoring, quantification of pro-inflammatory cytokines and myeloperoxidase activity, one can begin to assess these responses seen in inflammatory bowel disease.

Introduction to Titration

Source: Laboratory of Dr. Yee Nee Tan — Agency for Science, Technology, and Research Titration is a common technique used to quantitatively determine the unknown concentration of an identified analyte.1-4 It is also called volumetric analysis, as the measurement of volumes is critical in titration. There are many types of titrations based on the types of reactions they exploit. The most common types are acid-base titrations and redox titrations.5-11 In a typical titration process, a standard...

We demonstrate a protocol in which administration of the genotoxic agent azoxymethane (AOM) followed by three cycles of the pro-inflammatory agent dextran sulfate sodium (DSS) rapidly and consistently generates colon tumors in mice with morphologic and molecular similarities to those seen in human colitis-associated cancer.

Source: Vy M. Dong and Diane Le, Department of Chemistry, University of California, Irvine, CA This experiment will demonstrate a simple technique to titrate and obtain an accurate concentration of the organolithium reagent, n-butyllithium (n-BuLi). Organolithium reagents are extremely air- and moisture-sensitive and proper care must be taken to maintain the quality of the reagent so that it may be used successfully in a reaction. The n-BuLi titration experiments should be performed regularly...
