Fixed Tissue Immunofluorescence

Fixed tissue immunofluorescence is a microscopy-based method that uses fluorescently labeled antibodies to detect specific proteins or cellular structures in preserved tissue sections, making it valuable for studying tumor organization and molecular features. Tissue fixation stabilizes cellular architecture, while permeabilization and antibody staining allow probes to bind target epitopes; excitation light then induces fluorescence that reveals their location and distribution. In cancer research, this approach supports the evaluation of tumor markers, immune-cell infiltration, cell proliferation, and tissue microenvironment relationships. Multiplex staining can further compare several targets within the same section, linking molecular expression with spatial context and disease progression.

Background autofluorescence of biological samples often complicates fluorescence-based imaging techniques, especially in aged human postmitotic tissues. This protocol describes how autofluorescence from these samples can be effectively removed using a commercially available light emitting diode light source to photobleach the sample prior to immunostaining.
