Physiological Temperature

Physiological temperature is the temperature range at which an organism’s cells and biochemical systems normally function, typically about 37°C in humans, and it is essential for interpreting biological responses accurately. At this temperature, enzyme activity, membrane fluidity, protein folding, metabolism, and signal transduction operate within conditions that support cellular homeostasis; even modest deviations can alter these processes and trigger stress responses. In cancer research, maintaining physiological temperature in cell culture, organoids, and animal studies helps distinguish tumor-specific behavior from temperature-induced effects, while controlled temperature changes provide a framework for studying heat stress, treatment sensitivity, tumor metabolism, and the biological impact of hyperthermia.

In this paper we show a method for preparing acute brain slices in physiological temperature, using a conventional physiological solution without special modifications for the cutting (such as adding sucrose) and without intracardial perfusion of the animal before slice preparation.
