Telomerase Extension Reaction

The telomerase extension reaction is an in vitro biochemical assay that measures telomerase activity by tracking how the enzyme lengthens a telomeric DNA primer, a process central to chromosome-end maintenance and cancer biology. Telomerase uses its internal RNA template and reverse transcriptase domain to add repeated DNA sequences to the primer in the presence of suitable nucleotides and reaction conditions. In cancer research, this reaction helps quantify telomerase activity in tumor cells, compare activity across samples, and evaluate compounds designed to inhibit telomere maintenance. These measurements clarify how cancer cells sustain proliferative capacity and support the development of telomerase-targeted strategies.

In eukaryotic DNA replication, a single-stranded DNA fragment remains at the end of a chromosome after the removal of the final primer. This section of DNA cannot be replicated in the same manner as the rest of the strand because there is no 3’ end to which the newly synthesized DNA can attach. This non-replicated fragment results in gradual loss of the chromosomal DNA during each cell duplication. Additionally, it can induce a DNA damage response by enzymes that recognize single-stranded DNA.

Efforts to isolate the catalytic subunit of telomerase, TERT, in sufficient quantities for structural studies, have been met with limited success for more than a decade. Here, we present methods for the isolation of the recombinant Tribolium castaneum TERT (TcTERT) and the reconstitution of the active T. castaneum telomerase ribonucleoprotein (RNP) complex in vitro.
