Metabolite Identification

Metabolite identification is the process of determining the chemical structures and identities of small molecules produced, transformed, or consumed in a biological or chemical system. In chemistry, researchers typically separate metabolites by chromatography, measure their mass-to-charge ratios and fragmentation patterns with mass spectrometry, and use nuclear magnetic resonance or authentic standards to confirm structural assignments. These complementary approaches help distinguish related compounds, trace metabolic pathways, and characterize reaction products. Metabolite identification supports drug discovery, toxicology, environmental analysis, and systems biology by revealing biochemical changes, detecting unexpected metabolites, and linking molecular structure to biological or chemical function.

Nuclear magnetic resonance (NMR) spectroscopy is used to identify dysregulation in the metabolites in patients with various diseases. This technique allows the quantification of the deranged metabolites, unraveling the pathophysiological insights. Here, we describe the step-by-step procedure of the NMR-based approach for the metabolic characterization of the patients.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
