Reporter Accumulation

Reporter accumulation is the progressive buildup of a detectable reporter signal in cells, providing a record of gene activity over time rather than only a snapshot of current expression. In developmental biology, reporter genes are linked to regulatory sequences, and stable or slowly degraded fluorescent, luminescent, or enzymatic proteins persist after transcription, integrating repeated or transient expression into a cumulative signal. This approach helps researchers trace cell lineages, identify periods of pathway activation, and relate gene regulation to tissue formation and cell fate decisions. Reporter accumulation can therefore reveal developmental events that conventional instantaneous reporters may miss.

An Assay to Measure the Accumulation of a Fluorescent Antibiotic Probe in Bacterial Cells

This video demonstrates an assay to measure the accumulation of a fluorescent antibiotic probe in bacterial cells. Exposure to an efflux pump inhibitor disrupts the transmembrane proton gradient in bacterial cells, upon which a fluorophore-conjugated antibiotic is added. Inhibition of efflux pumps due to the disrupted proton gradient causes accumulation of the antibiotic inside the bacteria. Bacterial cells are lysed, and the fluorescence intensity is measured to confirm antibiotic accumulation.

The heterogeneous intra-tumoral accumulation of liposomes has been linked to an abnormal tumor microenvironment. Herein methods are presented to measure tumor microcirculation by perfusion imaging and elevated interstitial fluid pressure (IFP) using an image-guided robotic system.
