Hepatocyte Transfection

Hepatocyte transfection is a laboratory technique for introducing exogenous nucleic acids into liver cells, enabling researchers to alter or measure gene expression. Depending on the protocol, plasmid DNA, messenger RNA, or small interfering RNA crosses the cell membrane through lipid-based delivery, electroporation, or related methods, then directs protein production or gene silencing. In genetics research, transfected hepatocytes support studies of gene function, regulatory pathways, inherited liver disorders, and responses to candidate therapies. The method can provide transient or longer-lasting expression, although delivery efficiency, cell viability, and hepatocyte-specific responses must be assessed when interpreting experimental results.

An Introduction to Transfection

2023

Transfection is the process of inserting genetic material, such as DNA and double stranded RNA, into mammalian cells. The insertion of DNA into a cell enables the expression, or production, of proteins using the cells own machinery, whereas insertion of RNA into a cell is used to down-regulate the production of a specific protein by stopping translation. While the site of action for transfected RNA is the cytoplasm, DNA must be transported to the nucleus for effective transfection. There, the...

Gene Transfection toward Spheroid Cells on Micropatterned Culture Plates for Genetically-modified Cell Transplantation

This protocol describes a cell transplantation system using genetically modified, injectable spheroids. Cell spheroids are cultured on micropatterned culture plates and recovered after gene introduction using polyplex nanomicelles. This system facilitates prolonged transgene expression from the transplanted cells in host animals while maintaining the innate function of the cells.

Neuronal Transfection Methods

Transfection - the process of transferring genetic material into cells - is a powerful tool for the rapid and efficient manipulation of gene expression in cells. Because this method can be used to silence the expression of specific proteins or to drive the expression of foreign or modified proteins, transfection is an extremely useful tool in the study of the cellular and molecular processes that govern neuron function. However, mature neurons have a number of properties that make them...
