Brain Delivery

Brain delivery is the development of strategies that transport drugs, biologics, vaccines, or immune-modulating agents into the central nervous system, where access is limited by the blood-brain barrier. These approaches may improve exposure by using receptor-mediated transport, modifying therapeutic molecules, or packaging them in carriers that cross or temporarily alter barrier function while preserving neural protection. In immunology and infection research, brain delivery supports treatment of neuroinflammation, viral and bacterial infections, and central nervous system diseases by directing interventions to affected tissues. Improving delivery can increase therapeutic effectiveness, reduce systemic exposure, and enable more precise neurological research.

Convection-Enhanced Delivery of Antibodies into a Mouse Brain

2025

This video demonstrates a convection-enhanced delivery (CED) technique for antibody delivery into the mouse brain. The skull of an anesthetized mouse is exposed, and a burr hole is created at the target area for antibody delivery. A step catheter containing antibodies is inserted at the target site, and via an infusion pump, a pressure gradient is created to deliver antibodies at a controlled rate.

Intranasal Delivery of siRNA to the Mouse Brain Using a Positioning Device

Source: Ullah, I., et.al. A Positioning Device for the Placement of Mice During Intranasal siRNA Delivery to the Central Nervous System. J. Vis. Exp. (2019)This video demonstrates a precise intranasal administration technique for delivering neuron-targeting peptide-siRNA complexes to the mouse brain using a specialized positioning device. Anesthetized mice are secured in a head-down-and-forward position to optimize siRNA inhalation while preventing lung drainage. The targeting peptide...

Delivery of a Traumatic Brain Injury to a Mouse Brain

2025

This video demonstrates the delivery of Traumatic Brain Injury (TBI) in mice. After exposing the skull, the TBI site is marked. The mouse is then positioned under the TBI device. The impact is delivered by lifting the metal rod and allowing it to drop freely onto the impactor, which is in contact with the skull.

Microinjection-Based Delivery of a Therapeutic Agent into an Awake Rodent Brain

Source: Poland, R. et al. Rodent Brain Microinjection to Study Molecular Substrates of Motivated Behavior. J. Vis. Exp. (2015)In this video, a water-filled microinjection setup consisting of a syringe and microinjector tubing is used for precise drug delivery. A rat with a pre-implanted guide cannula undergoes targeted brain microinjection, enabling accurate administration of therapeutic agents in awake animals with minimal tissue damage.

Presented here is a protocol to transiently open the blood-brain barrier (BBB) either focally or throughout a mouse brain to deliver fluorescently-labeled antibodies and activate microglia. Also presented is a method to detect the delivery of antibodies and microglia activation by histology.
