Fecal Inoculum Preparation

Fecal inoculum preparation is the standardized processing of fecal material to create a reproducible microbial suspension for microbiology, immunology, and infection research. The process typically involves collecting a defined sample, homogenizing it in a suitable sterile diluent, and removing large particulate matter while preserving representative microorganisms under controlled conditions. Prepared inocula can support fecal microbiota transfer studies, colonization experiments, and ex vivo investigations of interactions between gut microbes and host immune cells. Consistent preparation is essential for comparing microbial communities, tracking infection or immune outcomes, and interpreting how intestinal microorganisms influence inflammation, barrier function, and disease susceptibility.

Adrenal activity can be assessed in the equine species by analysis of feces for corticosterone metabolites. The method offers a non-invasive option to assess long term patterns in both domestic and free ranging horses. This protocol describes the enzyme linked immunoassay involved and the associated biochemical validation.
