Luminol-based Assay

A luminol-based assay is a chemiluminescent analytical method that detects chemical or enzymatic activity by measuring light released from luminol oxidation. In a typical immunological application, hydrogen peroxide and a catalyst such as horseradish peroxidase convert luminol into an excited product that emits light, with signal intensity related to the amount of target or reaction activity. These assays support sensitive detection of antibodies, microbial antigens, inflammatory markers, and reactive oxygen species in infection research. Their broad dynamic range and compatibility with automated instruments make them useful for diagnostics, pathogen studies, and quantitative analysis of immune responses.

Chemical reactions can be classified as endothermic or exothermic depending on whether the reactions absorb or release heat when they change to products. The difference in energy between reactants and products is known as the enthalpy of the reaction. This is found by calculating the difference in energies between the products and the reactants, or ΔH. If ΔH is positive, the reaction is endothermic; if it is negative, the reaction is exothermic. While chemical reactions are traditionally...

The first step is a condensation reaction between 3-nitrophthalic acid and hydrazine. During this step, the carboxylic acid groups are substituted with NH groups to produce 3-nitrophthalhydrazide and two water molecules. You must work in a fume hood...

Please adjust quantities as needed. Before you get started, put on a lab coat, splash-proof safety glasses, and nitrile gloves. The chemicals that you will use are hazardous, so both solutions must be prepared and handled in a fume hood. Prepare 10 mL of a 10% by volume solution of...

An Assay to Detect Oxidative Burst in Arabidopsis Leaf Discs upon Immune Elicitation

This video demonstrates an assay to measure oxidative burst in Arabidopsis leaves upon immune elicitation. Leaf discs, cut from plant leaves, are incubated with bacterial peptides functioning as microbe-associated molecular patterns or MAMPs. Upon detection of the MAMPs, plant cells induce the production of extracellular ROS, which is detected using a chemiluminescence assay.
