Recombinant Respiratory Syncytial Virus

Recombinant respiratory syncytial virus (RSV) is an RSV engineered through molecular genetic methods to study viral biology, immunity, or vaccine performance. Using reverse genetics, researchers assemble modified viral genome components and recover infectious virus in cultured cells, allowing targeted changes to viral genes or regulatory sequences while preserving the virus’s replication cycle. These engineered viruses help clarify how RSV enters host cells, evades immune responses, and causes respiratory disease. They also support evaluation of vaccine candidates, antiviral strategies, and reporter systems that track infection, providing controlled tools for immunology and infection research.

We describe a method for generating and amplifying genetically modified respiratory syncytial viruses (RSVs) and an optimized plaque assay for RSVs. We illustrate this protocol by creating two recombinant viruses that respectively allow quantification of RSV replication and live analysis of RSV inclusion bodies and inclusion bodies-associated granules dynamics.
