Immunomodulatory Compound Identification

Immunomodulatory compound identification is the systematic process of finding and characterizing molecules that alter immune-cell activity, making it important for understanding host responses and developing therapeutic candidates. Researchers typically expose immune cells or biochemical targets to candidate compounds, then measure changes in signaling pathways, cytokine production, cell activation, proliferation, or inflammatory responses using screening and validation assays. In immunology and infection research, these studies can reveal agents that enhance protective immunity, suppress harmful inflammation, or influence pathogen-host interactions. Combining functional assays with target and pathway analysis helps distinguish specific immunomodulators from nonspecific toxic effects and supports the development of treatments for infectious and immune-mediated diseases.

A Fluorescence-Based Assay Using Engineered Lymphocytes for Screening Immunomodulatory Compounds

2025

This video demonstrates the screening of immunomodulatory compounds via fluorescent-based assay. Transgenic mouse-derived T lymphocytes with green fluorescent protein expression cassettes are treated with these compounds. The enhanced and reduced GFP signals enable the identification of the compounds' stimulatory and inhibitory effects, respectively.

This video demonstrates a method of converting dendritic cells (DCs) to tolerogenic DCs (TolDCs) using an immunomodulatory pharmacological agent. This conversion is confirmed by evaluating the TolDC function through their immunosuppressive effect on CD4+ T cells.

An In Vitro Assay for Evaluating the Immunomodulatory Impact of Monocytes on Leukocyte Proliferation

This video demonstrates an in vitro assay to explore the immunomodulatory influence of mesenchymal stem cells (MSCs) on effector T cell proliferation. The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

T-lymphocyte mitogenesis is accompanied by blastogenic transformation, whereupon the cell volume enlarges before cell division. Here, we describe a method to quantify blastogenesis in T lymphocytes using an automated cell counter with the capability of measuring cell diameters.
