Fc Receptor Phagocytosis

Fc receptor phagocytosis is the uptake and destruction of antibody-coated particles by immune cells, linking adaptive antibody responses to innate cellular defense. It begins when Fc receptors on phagocytes, such as macrophages and neutrophils, bind the Fc region of immunoglobulins attached to a microbe; receptor clustering then activates signaling pathways that reorganize actin and drive engulfment into a phagosome. The phagosome matures and fuses with lysosomes, exposing its contents to degradative enzymes and antimicrobial conditions. Studying this process clarifies host protection against infection, mechanisms of opsonization, antibody-based therapies, and inflammatory or autoimmune tissue injury.

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...

Measuring the Activation of Fc-Mediated Effector Functions by HA Antibodies

This video demonstrates an assay measuring the antibody-mediated activation of T cell effector functions. Adding an influenza virus hemagglutinin (HA)-specific monoclonal antibody to transfected mammalian cells expressing HA results in the formation of immune complexes with HA. Upon introducing engineered T cells expressing Fc receptors and a luciferase reporter, the T cells are activated by the antibody-HA complexes, and this activation is detected by adding a luciferase substrate and...
