Inhibitor Producer Colony

An inhibitor producer colony is a visible cluster of microorganisms that releases substances capable of suppressing the growth or activity of nearby cells, making it a useful indicator of microbial antagonism. The produced inhibitor diffuses through the surrounding culture medium and may create a clear zone or reduced-growth area in a susceptible indicator organism, depending on the compound and test conditions. In immunology and infection research, identifying these colonies helps characterize antimicrobial production, interactions within microbial communities, and potential biological control mechanisms. Such assays can support the study of pathogen competition and the evaluation of candidate inhibitory substances.

Here, we present a protocol to differentiate murine granulocyte-macrophage-colony-stimulating-factor-producing T helper (THGM) cells from naive CD4+ T cells, including isolation of naive CD4+ T cells, differentiation of THGM, and analysis of differentiated THGM cells. This method can be applied to studies of the regulation and function of THGM cells.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.
