Msc Leukocyte Co-culture

MSC leukocyte co-culture is an in vitro method that combines mesenchymal stromal cells with immune leukocytes to examine how these cell populations communicate and regulate inflammation. In controlled culture conditions, direct cell contact and soluble factors such as cytokines and chemokines can alter leukocyte activation, proliferation, differentiation, and effector functions, while leukocytes may also influence MSC behavior. In immunology and infection research, this model helps characterize immune modulation, inflammatory signaling, and responses to microbial challenges. Findings can clarify host defense mechanisms and support evaluation of MSC-based strategies for controlling excessive or dysregulated immune responses.

An In Vitro Assay for Evaluating the Immunomodulatory Impact of Monocytes on Leukocyte Proliferation

This video demonstrates an in vitro assay to explore the immunomodulatory influence of mesenchymal stem cells (MSCs) on effector T cell proliferation. The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

The therapeutic potential of mesenchymal stem/stromal cells (MSCs) is well-documented, however the best method of preparing the cells for patients remains controversial. Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.

Here, a protocol (MitoCeption) is presented to transfer mitochondria, isolated from human mesenchymal stem cells (MSC), to glioblastoma stem cells (GSC), with the goal of studying their biological effects on GSC metabolism and functions. A similar protocol can be adapted to transfer mitochondria between other cell types.

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.
