Neuraminidase Characterization

Neuraminidase characterization is the biochemical, structural, and functional analysis of neuraminidase enzymes, which remove terminal sialic acid residues from glycoconjugates and influence pathogen interactions with host cells. Researchers assess catalytic activity, substrate specificity, enzyme kinetics, stability, and inhibitor sensitivity using purified proteins, defined substrates, or cell-based assays. These measurements reveal how sequence and three-dimensional structure shape substrate recognition and catalytic efficiency. In immunology and infection, characterization helps compare neuraminidases from influenza viruses and other pathogens, evaluate antiviral compounds, and clarify their effects on mucus penetration, receptor release, and immune recognition.

A simple slot blot method was developed for the quantification of influenza viral hemagglutinin and neuraminidase using universal antibodies targeting their most conserved sequences identified through bioinformatics analyses. This innovative approach may provide a useful alternative to quantitative determination of all viral hemagglutinin and neuraminidase.
