Nickel Affinity Column

A Nickel Affinity Column is a chromatography tool used to isolate proteins engineered with a polyhistidine tag, making it valuable for producing purified molecules for immunology and infection research. The column contains immobilized nickel ions that coordinate with histidine residues on the tagged protein, while unwanted proteins pass through during washing; imidazole is then added to compete for nickel binding and elute the target protein. This method supports the preparation of recombinant antigens, enzymes, and other proteins for antibody production, immune assays, structural studies, and investigations of pathogen biology. Its selective binding and straightforward workflow enable reproducible protein purification from complex cell lysates.
