Polyclonal Antibody Fragments

Polyclonal antibody fragments are antigen-binding portions of antibodies collected from multiple B-cell clones, retaining recognition of several epitopes on the same antigen. This molecular diversity can improve target coverage in immunology and infection research. Enzymes such as papain or pepsin cleave antibodies to generate Fab or F(ab′)2 fragments; these fragments preserve variable-region binding while removing much or all of the Fc region and its associated effector functions. Their smaller size and reduced Fc-mediated interactions support applications including pathogen detection, antigen localization, neutralization studies, and diagnostic assay development, while helping researchers examine antibody specificity and targeted binding.

We provide a method to simultaneously screen a library of antibody fragments for binding affinity and cytoplasmic solubility by using the Escherichia coli twin-arginine translocation pathway, which has an inherent quality control mechanism for intracellular protein folding, to display the antibody fragments on the inner membrane.

Here, we selectively target antibodies against a specific member of a highly conserved family of proteins by immunizing animals with their most divergent regions followed by removing cross reactive antibodies by pre-adsorption.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.
