Tolerogenic Dcs

Tolerogenic dendritic cells (tolDCs) are antigen-presenting cells that promote immune tolerance rather than inflammatory activation, helping prevent harmful responses to self or harmless antigens. They process and present antigen under conditions of limited costimulation and increased regulatory signaling, which can induce T-cell anergy or deletion and support the expansion of regulatory T cells. In immunology and infection research, tolDCs help explain how immune responses are restrained after antigen exposure and how pathogens may influence immune regulation. Their properties also inform experimental approaches for treating autoimmune disease, improving transplant tolerance, and designing antigen-specific immunotherapies.

Generation of Immature, Mature, and Tolerogenic Dendritic Cells from Monocytes

This video demonstrates the generation of immature, mature, and tolerogenic dendritic cells, or DCs, from human monocytes. The incubation of monocytes with growth factors and cytokines leads to the production of immature DCs. In response to immune regulatory molecules, immature DCs differentiate into tolerogenic and mature DCs.

Immature dendritic cells can be selectively differentiated into tolerogenic or mature dendritic cells to regulate the balance between immunity and tolerance. This work presents a means to generate from immature monocyte derived dendritic cells (moDCs), in vitro tolerogenic and mature moDCs that differ in metabolic phenotypes.

We describe a procedure to assess the capacity for pharmacologic agents to generate tolerogenic dendritic cells from naïve monocyte-derived dendritic cells in vitro and validate their potency by autologous regulatory T cell generation.
