Advanced Precancerous Lesions

Advanced precancerous lesions are abnormal tissue changes that show substantial progression toward invasive cancer but have not yet breached the tissue boundaries that define malignancy. They typically arise as accumulating genetic and epigenetic alterations drive dysregulated cell growth, impaired maturation, and clonal expansion, producing high-grade dysplasia or related changes detectable by histopathology and, in some organs, imaging or endoscopy. In medicine, recognizing these lesions supports risk stratification, targeted biopsy, surveillance, and removal or local treatment before invasion develops. Studying them also clarifies early cancer evolution and helps evaluate prevention strategies and biomarkers for progression.

Laser microirradiation is a useful tool for studies of DNA repair in living cells. A methodological approach for the use of UVA lasers to induce various DNA lesions is shown. We have optimized a method for local microirradiation that maintains the normal cell cycle; thus, irradiated cells proceed through mitosis.

Sectioning Mammary Gland Whole Mounts for Lesion Identification

We developed a method to successfully remove, process, section, and stain, for histopathological evaluation, mammary tissue that had originally been fixed on slides as whole mounts. This method may promote the collection and evaluation of mammary gland whole mounts in reproductive and developmental test guideline studies.

A protocol for performing unilateral 6-OHDA lesions of the medial forebrain bundle in mice is described. This method has a low mortality rate (13.3 %) with 89% of the surviving animals showing >95% loss of striatal dopamine and 90.63±-4.02 % ipsiversive rotational bias towards the side of the lesion.
