Allograft Survival Model

An allograft survival model is an experimental system used to measure how long transplanted tissue or an organ from a genetically different member of the same species remains functional, providing a direct way to study transplant rejection and tolerance. The model typically involves transplanting donor tissue into a recipient and monitoring graft function, appearance, or survival over time as immune cells recognize donor antigens and initiate inflammatory or destructive responses. Researchers use allograft survival models to evaluate immunosuppressive therapies, define mechanisms of alloimmunity, and identify factors that promote long-term graft acceptance. These findings support the development of safer strategies for transplantation medicine.

Here, we present a protocol to study the immunology of rejection. The surgical model presented reports a short operating time and a concise technique. Depending on the donor-recipient strain combination, the transplanted kidney may develop acute cellular rejection or chronic allograft damage, defined by interstitial fibrosis and tubular atrophy.
