Enteric Neuronal Loss

Enteric neuronal loss is the reduction or destruction of neurons in the enteric nervous system, the intrinsic neural network that coordinates gastrointestinal motility, secretion, and blood flow. It can arise when inflammation, ischemia, infection, toxins, or other insults trigger neuronal dysfunction, apoptosis, or degeneration; the resulting reduction in neural signaling disrupts communication between enteric circuits, smooth muscle, and intestinal epithelial cells. In medicine, enteric neuronal loss is investigated as a mechanism underlying dysmotility, constipation, intestinal pseudo-obstruction, and some inflammatory or neurodegenerative gastrointestinal disorders. Measuring neuronal density and associated changes in motility can support disease classification, treatment research, and development of neuroprotective or regenerative strategies.

We demonstrate a cell culture protocol for the direct study of neuronal and glial components of the enteric nervous system. A neuron/glia mixed culture on coverslips is prepared from the myenteric plexus of adult mouse providing the ability to examine individual neuron and glia function by electrophysiology, immunohistochemical, etc.
