Hemagglutination Inhibition Assay

The hemagglutination inhibition assay is a serological method that detects and estimates antibodies capable of preventing viral hemagglutination, a reaction in which viral surface proteins cause red blood cells to clump. In the assay, patient serum is mixed with a virus or viral antigen, followed by red blood cells; if specific antibodies bind the viral hemagglutinin, they block attachment to the cells and prevent visible agglutination. The resulting inhibition pattern or antibody titer helps assess prior infection, immune response, or vaccine-induced protection. Used in medicine and virology, this assay supports influenza surveillance, serological diagnosis, vaccine evaluation, and comparisons of antigenic variation.
