Human Tissue Processing

Human tissue processing is the preparation of tissue specimens for microscopic examination, diagnosis, research, and long-term preservation. The process typically stabilizes tissue through fixation, removes water by graded dehydration, clears it with an intermediate solvent, and embeds it in a supportive medium such as paraffin, allowing thin sections to be cut and mounted on slides. Subsequent staining highlights cellular and extracellular structures for evaluation by light microscopy, while specialized processing can preserve molecular targets for immunohistochemistry or molecular analysis. In medicine, reliable tissue processing supports cancer diagnosis, disease classification, treatment planning, pathology research, and the development of tissue-based diagnostic methods.

Processing of Human Reduction Mammoplasty and Mastectomy Tissues for Cell Culture

A method to process human mammary surgical discard material is described. Processed tissue, in the form of organoids, can be stored frozen indefinitely or placed in culture for long-term growth. This method enables experimental examination of normal human epithelial cell biology, and the effects of exogenous perturbations.

Translational cancer research is dependent on extraction of human tissues. Much work has gone into optimizing extraction methods for ex vivo analysis. Here, we describe tissue processing methods allowing for maximal data output from limited samples.

This protocol demonstrates a reliable and effective technique to isolate primary fibroblasts from either premenopausal or postmenopausal human vaginal tissue. Existing protocols for vaginal fibroblast isolation do not consider the challenges of cell isolation from senescent tissue. Vaginal tissue was obtained from women after pelvic organ prolapse surgery.

Tissue Engineering of a Human 3D in vitro Tumor Test System

Methods to create human 3D tumor tissues as test systems are described. These technologies are based on a decellularized Biological Vascularized Scaffold (BioVaSc), primary human cells and a tumor cell line, which can be cultured under static as well as under dynamic conditions in a flow bioreactor.

Electrospun scaffolds can be processed post production for tissue engineering applications. Here we describe methods for spinning complex scaffolds (by consecutive spinning), for making thicker scaffolds (by multi-layering using heat or vapour annealing), for achieving sterility (aseptic production or sterilisation post production) and for achieving appropriate biomechanical properties.
